Increased somatic mutation burdens in normal human cells due to defective DNA polymerases


The higher mutational load does not appear to lead to the early onset of the signs and features that we usually associate with aging.


People with POLE / POLD1 germline mutations do not show clear signs of premature aging. These results do not support the model in which aging is attributed to cellular malfunction resulting from the burden of somatic mutations accumulated throughout life.
